Usage recommendations for probe T3-CLK and control T3-CLK-N

In cells

Use at concentrations < 100 nM to avoid off target effects and on target cytotoxic in longterm experiments. In case one wants to see the strongest effects of T3-CLK on alternative splicing, higher concentrations can be used, but cytotoxicity and G2/M cell cycle arrest will occurre after approximately 20 h.

The CLK probes, MU1210 and SGC-CLK-1, available from the main SGC website, can be used as additional control to dissect CLK activity further.

Use the recommended concentration to avoid off-target toxicity and/or off-target activity. Using concentrations that are higher than recommended could lead to false positive or confounding results.

In vivo

Not tested in any in vivo model.
