Usage recommendations for probe WEB2086 and control WEB2387


In cells

Use at concentration of 300 nM.

Use the recommended concentration to avoid off-target toxicity and/or off-target activity. Using concentrations that are higher than recommended could lead to false positive or confounding results.

In vivo

Tested in guinea pigs with 0.1 - 2 mg/kg po and 0.01 -0.1 mg/kg iv dose. Determination of the recovery of respiratory flow and mean arterial pressure (measure of hypotension)) in guinea pigs: WEB2086: Respiratory flow: ED50 = 0.07 mg/kg (po), ED50 = 0.018 mg/kg (iv); Mean arterial pressure: ED50 = 0.066 mg/kg (po), ED50 = 0.016 mg/kg (iv). WEB2387: Respiratory flow: ED50 = 1.55 mg/kg (po), ED50 = 0.081 mg/kg (iv); Mean arterial pressure: ED50 = 1.2 mg/kg (po); ED50 = 0.086 mg/kg (iv). WEB2086 reduces bronchoconstriction, hypotension, microvascular leakage, and anaphylactic shock amongst many others as shown in various animal models.
